Solid phase extraction is one of the most widely applied sample preparation techniques in analytical chemistry, used to isolate, concentrate, and clean up target analytes from complex matrices before chromatographic analysis. The selectivity of the SPE sorbent — whether reverse phase, ion exchange, mixed mode, or specialty formats like immunoaffinity or molecularly imprinted polymers — determines how effectively interferences are removed and how completely the target analyte is retained and recovered. Method sensitivity, matrix effects in LC-MS detection, and the ability to meet reporting limits in regulated methods are all directly dependent on the performance of the SPE format selected. Cartridge bed mass, sorbent particle size, and housing dimensions must also be matched to the sample volume and vacuum manifold system in use to ensure reproducible flow rates and consistent recoveries across an entire extraction batch.
